Mike Berridge completed a doctoral degree in cell biology at the University of Auckland in 1971 exploring the mechanism of action of plant growth hormones. Following postdoctoral research in developmental molecular biology at Purdue University, USA, and experience as a staff scientist at National Institute for Medical Research, Mill Hill, UK, he returned to Wellington in 1976 as the second Malaghan Research Fellow where he established the Cancer Cell & Molecular Biology Research Group and was a founding scientist of the Wellington Cancer & Medical Research Institute, later renamed the Malaghan Institute of Medical Research in recognition of major personal support by Len and Anne Malaghan.
Mike Berridge received a James Cook Fellowship in the health sciences in 2003 and was awarded the Health Research Council Liley Medal for outstanding research on cellular metabolism in 2016. In the same year he was a semi-finalist in the KiwiBank New Zealander of the Year Awards.
Prof Berridge’s current research interests include cancer cell energy metabolism and mitochondrial gene transfer between cells in human disease. As a science communicator, he recently published “The Edge of Life” (2015), and “Sugar, Rum and Tobacco: Taxes and Public Health in New Zealand” with Lisa Marriott in 2017.
Our major scientific discovery that mitochondrial genes move to tumour cells without mitochondrial DNA has challenged a central dogma in cell biology that genes are constrained within cells and partition only during cell division. Acquisition of mitochondria from normal body cells restores tumour forming ability in breast cancer and melanoma models by enabling mitochondrial respiration.
This discovery has opened doors that will drive our research programme in new directions over the next decade. While exploring the difference in gene expression between breast cancer cells with and without mitochondrial DNA, we discovered that the mitochondrial genome is required for expression of several nuclear immune and stress response genes. Although similar stress response genes were observed in our melanoma and brain tumour models, the immune response genes were tumour type specific. This novel finding will allow us to determine how different tumours have evolved to hijack immune cells and other normal tissue cells for their own benefit, allowing tumour progression, immune escape and metastasis. This new research frontier has identified new targets for the development of novel cancer therapies, and is anticipated to lead to improved cancer immunotherapy.
We have found that mitochondrial transfer between cells also occurs in a brain tumour model raising profound questions about how the brain manages energy production in the central and peripheral nervous systems. Using our brain tumour model we will explore mitochondrial movement from normal brain cells to brain tumour cells and between normal brain cells. We will also determine how developing brain tumours control anti-tumour immune responses, a field that is in its infancy.
Since 2011, the discovery culture of the Cancer Cell Biology Group has become increasingly an internationally collaborative venture with Prof Jiri Neuzil, who holds positions at Griffith University, Gold Coast, and BIOCEV Biotechnology Institute, Prague West, and with Dr Lan-feng Dong at Griffith University. These collaborations have resulted in several high impact and highly cited publications, the most recent involving 15 research Institutions worldwide from eight different countries. In Wellington our small group collaborates with Dr Melanie McConnell and her team at Victoria University, with Associate Professor Patries Herst, Radiation Therapy, University of Otago Wellington, and internally with Malaghan Institute scientists whose dendritic cell research has highlighted dramatic mitochondrial gene involvement.
The main focus of the Cancer Cell Biology Group’s research is discovery. It applies new knowledge and methodologies to cells, cell systems, and complex tissues and organs to the benefit of human diseases, with a particular emphasis on cancer and diseases involving altered energy metabolism.
Our vision is that better understanding of tumour cell biology, brain inflammation and anti-tumour immune responses will generate new knowledge that will lead to better management and treatment of cancer and the major neurological diseases, and enhance the well-being of our ageing population.
